(i.e. Requiring the lowest ## of materials and shortest time to build) Do I simply need to skill everything that the blueprint requires to 5? Or is there some base level of skills that is independent of blueprints that does this? (i.e. Industry 5/Science 5 make you a perfect builder)

To get the lowest possible build requirements, the only skill you need is Production Efficiency to 5.
Lowest time - Industry 5 and Cybernetics 4 (For the manufacturing speed implant)

To be a perfect builder you also need a hangar full of minerals and a market to sell your stuff with profit and a set of fully researched blueprints, and slots for building, souveranity for building big stuff and protection against loosing your stuff while you build it. You also need ships for transport (freighter or jump freighter, cloaked industrial ships, rorqual etc.)
Its not only skills and isk to be a perfect builder.

Cyber 5 for the F60 and then the ability to use a POS for faster production [minerals costs go up though] --

Also, while T1 manufacturing skill requirements are awfully easy, if you're going to start T2 manufacturing you are going to need a lot of Science skills (and perhaps PI skills to create some T2 components).
