ok, those come about 16 normal missions and you get nice loot and/or faction standing increase, but do you also loose standing? e.g. if i do galente storylines would i loose amarr and caldari standing? can i safely turn the mission offers down before i accept it? what would i loose in term of possibilities and later options if i turn them down?

Yes, you take a larger hit from what I have heard when you do storyline missions for the factions that you are against.
Turning down a storyline will do two things, it will lower your standing with the storyline factions standing and raise the faction standing for the ones that you were against.

turning down the mission (the old i don't have time for this reply) is ok last i checked but failing the mission does what the first reply mentioned..
turning down - np accepting then failing - faction hits and boosts accepting then succeeding - faction boosts and hits

When you do a "storyline - important" mission, you gain X points of standing toward the faction that gave you the mission, X/2 toward the allied faction (i.e. if you do a Gallente storyline, you'll gain X in Gallente Federation and X/2 in Minmatar Republic), BUT you also loose X/2 toward the "main enemy" of the faction (Caldari in this case) and X/4 toward the "secondary enemy" (Amarr here).
The losses are less than the gains, so you can, with a lot of work, raise your standings toward all four factions ... but they even out at around 5.0 while doing that, no way to reach the 8.4+ needed for the Navy BPCs given by some COSMOS agents.
I've been moving between Caldari and Gallente agents for the last year or so, in an effort to raise all my faction standings evenly ... the pirate factions hate me, but since I'm an empire carebear by choice, that doesn't bother me

Have a look at http://www.newedenlibrary.net/eon/standings.shtml for details of how standing changes work. The table found at http://www.newedenlibrary.net/eon/faction_standings.shtml shows full details of the +ve and -ve standing changes on a per faction basis.

This has been my experience. And, in a multi-part mission, the failure penalty is extreme. I took a 5% hit once. Ouch. Phoenix Logistics Industries |
