I know you can't harvest moon minerals in 0.4 systems, but is it possible to run reactions at a POS??

The attribute value restricting use of reactor arrays and moon harvesting arrays is identical - where you can't use one you can't use the other either.
So, no, reactions are not possible in 0.4, just 0.3 and below.

More question marks definitely make the question more important.

True, but there is a stacking penalty involved. After about four or so question marks the diminishing returns pretty much minimizes the value in adding any more. If you have the calibration points left for it you can sometimes fit a few '!' or '*' which can squeeze out bit more importance in certain situations.
