It's brighter and has significantly more mass, but it's much smaller in size (VY Canis Majoris 600-2600 solar radii vs. R136a1 35,4 solar radii).

As an astronomy student, I have to say, if these stars are really as massive as they currently estimate, it would go against most everything I've been taught about stellar evolution/structure. It's currently generally believed that stars with masses more than about 100 to 150 solar masses can't form. Simply because the pressure from the light they produce* would more than counter the stars own gravity.
I've just read thourgh part of the scientific paper and the highest mass they mention is the initial mass, it's 20-25% lighter now. Stars this massive will lose a lot of mass quickly and die quickly as well (few mil years, maximum).
Will be interesting to see if other scientist will find similar estimated for this star. Because I can only imagine some people will want to check these estimates for themselves, since this could change our understanding of stellar formation/structure/evolution.

The radius of a star does not only depend on it's mass, it depends on its age as well. Old stars have a habit of expanding enormously just before dying. Going from a few times the size of the sun to the size of earth's orbit (the sun will do this, roughly) to filling up something the size of Uranus' orbit, maybe more. In general: heavier = bigger and older = bigger.
*Light (or more correctly electromagnetic radiation) can exert pressure, which is simply called radiation pressure.
