I'm about to begin grinding Level 3's in order to access this Level 4 R&D agent. I have all the skills needed to do R&D with the agent so that isn't a problem. The agent is of good quality (Q8 I think) and my skills are a mix of 4's and 5's (5's being the ones that are required).
My question is that how many points does it require to obtain a datacore and how many points could I make in a day (or hour? Week?) or so? This will be my first time using an R&D agent so I was looking for ball park figures on what I can expect?
Is it worth the short grind to get R&D agents? Or am I better off just buying the datacores off the market?

The RP cost per datacore varies based on the datacore. But the short of it is that a level 4 agent (especially Q8) will earn you more RP per day than the cost of 1 datacore. And that is without doing the offered bonus missions.

I think most (if not all) datacores require 50 or 100 RPs; however, the agent produces twice as many RPs per day for the 100 RP datacores as the 50, so it comes out as a wash. LvL 4 agents will probably earn you somewhere around 1.5 datacores per day (double that if you do their piddly missions).

I was grinding faction status to get my R&D agent status up, so it helps in multiple ways if you are an industrialist - refining tax decreases, datacore increases, broker fee reductions, POS anchoring, etc., so it may seem to be a bit of a waste to grind strictly for datacores, there may be additional benefits. Finally, by running the R&D agent missions, you can boost your storyline missions considerably.
I like to make a few types of datacores for invention, not just for the savings but for the convenience - I use agents near my manufacturing hub.

Regarding their missions, are they required? The agent I'm looking at is about 10 jumps away. Not TOO far but a hassle if I had to do one for him a day. Would I lose standings if I rejected them? Could I just let them expire or would I have to go there and reject them?

A more or less recent change made it so you finally CAN gain standings with research agents by doing their missions. Gaining standings means higher effective agent level, which means more RP.
P.S. All missions are optional. You don't lose any standings if you don't bother with them. However, it would be good to run at least one, to make the connections skill kick in. The reward is also one day's worth of RP, the missions are usually trivial, and they also count towards storyline missions, so, you know, if you're around, why not just run them. _

The social skills that change effective standings no longer apply to base null standings entities. In other words, even if you have Connection L5 trained, if you have no standings with an agent, the effective standings are still zero, but if you run even the lousiest mission and go up to +0.01 standings, your effective standing jumps to +2.01 _

Isn't the standing applied when you ask the agent to do some research for you? I always assumed it was, but never checked. I have 15 research projects going across 3 characters and have never done a research mission for any of them .

I just tried running a mission to see what effect it had.
Pre mission: 0 standing and 152.32 datacores per day
After mission 0.07 standing (connections 3 raises it to 1.26) and 153.60 datacores per day.
Obviously with better connection skills you'd get more of a rise than I did but I wouldn't rush out to run missions for such a small reward.

You mean RP points, not datacores :p.
I take it that's a starship engineering research project?
