That said, if you're just looking for a way to quickly calculate individual item profit without regard to actual sales you've made yet, I think you could probably make a simple spreadsheet calculator.

As a first step - a calculator is just fine.
Fees are known - you've got between 0.5 and 1% sales tax dependent on skills. And you've got c. 0.3 - 1% brokers fees on the sale and purchase dependent on skills and standings.
So, worse case scenario, and to allow a little wiggle room (though not much if the market shifts) - call it 1% tax and 2% brokers fees (1% each for buy and sell). So that's 3%.
Take your buy price. Multiply by 1.03. Ensure sell price is higher than this figure.

Minor Tweak: 1% of that is based on the buy and 2% on the sell. Since the sell is higher than the buy, you should take your sell, subtract 3% and have to be lower than that. Not much of a difference unless you're working with really close margins.

On the open market, "fees" have a full range between 0.5% and 9% overall, that's the minimal and maximum ranges. Note here I said "fees" have a minimum of 0.5%, this is selling directly to Buy/Bid Orders. A 9% fee would be -10 Faction -10 Corporate standing with the corporation you're placing the orders at, you place a timed buy order to acquire the items, you also place a timed sell order to sell the items, and have no skills.
As a player with No standing and no skills will have between 1% and 3% depending on how they acquired and sell the items.

Chuckle, yep, which is why I said "c. 0.3-1%". Yes it could go outside those ranges. I took a punt on the OP's likely position, rounded up for a bit of wiggle room and gave a rough calculation as a starting point.
With the greatest of respect to the OP, he struck me as someone in need of a quick and easy answer as a starting point, an introduction. Throwing figures to umpteen decimal places at (someone who struck me as) a novice - well I didn't figure that would be helpful. Start out with the basics and explain how those figures are arrived at so the learner can work out more indepth and personal calculations :)

check out Eve Nexus I don`t think its suported much now, but you get a handy graph and, not so much a calculator, but it breaks down the broker fees and taxes using your API and gives you your final profit for each item. Its not to difficult to understand how to use either...Uses Java, but fails miserably with x64 version installed.

It does take the brokers fee and tax into consideration. You can check it by hovering the mouse over the ISK/profit column as it will show the fee and tax there (and those are calculated into the profit).
