I've been playing Eve for a week so the variety of ships I can pilot is pretty limited. However, for level 2 missions I've been having good luck with my Gallente Destroyer. I thought I might show my current configuration and elicit some advice on what to replace, if anything. For the moment this is strictly an anti NPC ship.
High: 4 125mm Railguns 1 Neutron Blaster 1 Ion Blaster
* all with Antimatter ammo
Mid: 1 Afterburner 1 Capacitor Recharger
Low: 1 Armor Repairer 1 Diagnotic System 200mm armor plate
This ship seems to do well against NPC ships, but big mobs of frigates often force me to warp away to recover. How can I tighten up my load-out, or make use of different tactics with a slow ship with many high slots?
Or maybe I should just go for a Cruiser as soon as I can...

I have a Gallente destroyer myself, but I have lots of skills to help me fit better stuff. This is what I had on it:
Hi: 8x Light Ion Blaster IIs (you should use Is) Med: 1x 1MN MWD (training for this is a must) 1x Small Shield Booster I Low: 3x Capacitor Relay Is
Think thats what I had, works pretty well. MWD right up to them and blast them in 1 volley. Insane damage, but requires a MWD. ********** Everyone deserves to die. You go first.

When you say "you should use Is" do you mean tech I guns or Ion blasters? [EVE players love to abbreviate, don't they?]
I'm several days away from the MWD since it takes two level IV skills but it gives me something to work on. I've mostly been concentrating on the array of Gunnery skills you need to hit the broad side of a barn.
Shield tanking, huh? I thought the plate and the armor repairer worked well. Don't you feel fragile against groups of five or six frigates?

I haven't flown a destroyer specifically, but can give some tips.
If you can (and I hear you probably cant because of powergrid issues) fit 150mm guns. Use a longer range ammo.. Something to make your optimal around 15km is good.
Dont bother with blasters - the point of the config Im suggesting is to stay out of the range of the npc frigates.
Your mid looks fine.
In your low slots, keep the armor repper just in case ya run into trouble. Dump the armor plate. Depending on what you need, you want either coprocessors (unlikely), Reactor Control Units (more power), Power Diagnostic systems (a little power and some cap recharge/shield recharge), or damage modifiers.
Try to aim for damage mods but if you need the RCU/PD to fit the extra gun (or other damage mods) then throw one in.
When you warp in, make sure you keep your range from the nearest npc at 15km all the time. With the AB, you should have no trouble doing this. Most level 2 npcs arent very fast at all.
You could also try to fit a full on short range config, but I hear destroyers dont tank very well at all.
In any case, against npcs its generally better to specialize than mix gun types like that (until you start fighting higher level npcs whom you no longer have complete control over the combat situation).

Getting in close does have the virtue of being nearby all those containers - a time consuming chore, however lucrative it may be.

Getting in close also makes you dead against any serious number of NPCs.

Dirus, your config looks like a good config for anti frigate duties, but will not work very well for level 2s. Destroyer can not tank well enough to do level 2 missions up close. Thorax or Vexor might be able to do them close, but not a destroyer.
Druuga, stick 8 biggest guns you can that give you longest range (125mm rails are minimum, you can fit them with no skills, but get 150mm as soon as you can and replace 125mm with them as much as your PG allows) and you will pop enemies in 1 volley while the only tanking you will have to do is missles.
setup something along those lines:
8 125mm --------------- ab,cap recharger (can be replaced with web later once you have good energy skills) --------------- armor repairer, 2 cprs
pretty good tanking setup. can hit far and hard while easily tanking several missle frigates at once.
or
8 125mm ---------------- ab,shield booster ---------------- 3 power diags
second config gives you a lot more PG so you might be able to replace many 125mm with 150mm. Not as good tanking ability as first one but longer range.

I personally prefere to keep out of range of enemy guns myself, so I quickly learnt Caldari Frig. 3 and mainly use their Destroyer since it gives bonuses to optimal range. (Doing learning skills now, so Cruisers are still a few weeks off)
This worked perfectly untill the missile "nerf" with the last patch. Now since missiles shoot further then my locking range and defenders don't work, I have to pass missions where I have to fight multiple missile opponents at once. For single missile opponents (frigs) I take out my Gallete Destroyer and go for close range/high damage.
Fittings:
Hi: 3x 150mm Prototype Gauss Guns 1x 150mm Carbide Railgun 1x 125mm Prototype Gauss Gun
Mid: 1x Small Shield Booster 1x Monoprop. Hydra. Booster (Afterburner) 2x Micro Shield Extenders
Low: 1x Auto Gain Tracking Enhancer I (+11% range on weapons)
With Destroyer 2 and Sharpshooter 1 I have my typical killing Zone at 23km. (Loading Irid., Tungst. and Iron in the 3 weapon types) but can go up to my locking range by changing ammos, which can be really good against some cruisers during level 2 missions. Afterburner typically keeps all opponents at bay (except those that use MWD).

If I were to start over again, that is exactly what I'd be doing. Destroyers are extremly fragile and not too good at tanking. The large number of guns is "nice" but with the 25% rof penalty, they are equal to a ship with 6 guns, but cost more in fitting and in isk (for the guns).

Taketa De, micro shield extenders are not good, you will be much better off by replacing them with 1 shield booster amp and 1 cap recharger. It will give you a lot more survivability then those extra shields.
