It's known that you will be given a storyline mission every 16 normal agent missions...
Does this mean after 16 L4 Agent missions I receive an L4 quality storyline mission? 16 L3 missions to a L3 quality storyline? L2...etc.?
Does the NPC corp I use have any effect - i.e. can I do 16 L4 missions from 5 different corps and then get the storyline mission?

It's anywhere from 10 to 16 agent missions before you pull a storyline. The last mission you do determines the difficulty of the S-line mission. Doesn't matter from which corp you do them.

It is now - and has been for the longest time - 16 sharp. The storyline mission agent will pick from the mission pool for the appropriate level.

that is 16 with the same agent.
you must do 16 with the same agent, though when you do them doesn't matter. if i go back to my lvl1 agent now, and run a few missions, i will likely get a story mission sooner than 16 because of missions i have left that i never completed the 16 for

Yes, one important mission every 16 missions.
I can not confirm that storyline agents pick from the mission pool with similar difficulty.
I was running L4 mission for a -19 quality agent in a 0.3 system and had good important missions aka infiltrate the enemy base, double agent, shipyard theft which gave 8% and more faction standing boost.
Now I am running L4 missions for a +20 quality agent in a 0.1 system and I receive important missions like outbreak and three times in a row important personal transportation. That's very diappointing because they only give a faction boost below 2%.
Do you think it's the 'wrong' storyline agent?

I had 2 Angel strike missions with a 3 normal mission gap, and i only use one kill agent. .................

When building my Amarr Navy faction I used 3 - lvl 2 agents at the same time all of them worked for Amarr Navy. I did not have to do 16 sets of missions to get a story line mission and I did not get 3 story line missions (1 from each agent). I got 1, and that 1 agent mission came very quickly. Also the agent that I hit the 16th mission on determined the corp that my story line mission was from.
I will explain how this was tested. I sometimes got story line missions for Court Chamberlin, and sometimes Sarum Family. By the time I had worked my way up from a lvl 2 Quality 5 agent to a lvl 3 qaulity 18 agent, I had done somewhere around 2 story line missions for Court Chamberlin and 4 for Sarum Family. So the randomness in who I got assigned to was determined by who the 16th mission was for, of the 3 agents I was working for.
To further prove this, my friend was building his faction and only worked for each agent until he got a story line mission. 2 of those agents gave Court Chamberlin story line missions, and 1 of them gave Sarum Family. I have since tested this and found the same to be true.
Ministry of Internal order use storyline agents from the same corp, which I have also tested and confirmed this with other people. This holds true for the lvl 2 agents and the lvl 4 agents in Penirgman. I have not done enough missions for the lvl 3 agent to see if it also uses an agent within the same corp. So in turn building faction with Ministry of Internal order is MUCH faster. 1 day of missions got me access to a high quality lvl 3 agent.

Short version. From my experience 16 missions within the same corp reguardless of agent gets you a storyline mission, the agent that you complete that 16th mission for determines what corp the story line agent will work for.

I've not been doing missions that long and I have a question that some of you may have come across before.
I am doing lev 1 missions for a +13 Minmatar Mining Corp Agent. So far I have had two storyline missions but the puzzling thing to me is that, the storyline agent is from the Republic Military School and completion of this mission doesn't alter my standing to Min Mining at all. In fact, the last storyline mission didn't seem to affect any standings at all.
With what appears to be an average 0.01 standing increase per normal mission, I need all the help I can get from the story ones.

Storyline impacts the standing towards the base agent's faction, not his corp.

No, you receive corp and faction standing for an important mission.
The storyline agent must not come from the same corp you've previously done your 16 missions for, though.
