I have an alt in a one man corp, trying to work towards a high-sec POS. He has only started doing missions recently, but he already has corp/faction standing, but the corp he is in has none whatsoever.
So how is this calculated? and what do I have to do to increase the corp standings?

Im pretty sure your corp gets some insanely low standing increase every time you do a mission. Say the agents standing goes up 20%, the agents corp's standing goes up by 3%, and the agents corp to his corp standing goes up by 1%. don't quote me on the numbers though. ____________________ (\_/) (0.o) (>.<) Bunny has been revived. Does the number of members in the corp make a difference?

As far as I know, yes, the number of players that ATTEMPT a corp mission can have a huge impact on its standings for the corp, and I think in return that has an impact on your faction standings also. You can have 20 members, but if only 2 do Caldari Navy, then only the 2 people will count towards the corps standing in Caldari Navy... however, all 20 members play a part in the corps entire faction standings.
Example... someone has Caldari Navy to 8.0 standing, someone in your corp decided to try 1 mission, hates it, and doesnt do it again... well... he just cut your corps standing in half... if only those 2 people in the corp have done Caldari Navy missions.
Again, this is just from the numbers I have looked at... not exactly sure.
