AFAIK...
1. Region boundaries affect trading the most. 2. The factions (Caldari, Minmatar, Gallente,...) claim sovereignty in terms of regions. 3. Player alliances usually claim territory in terms of regions.
4. For constellations, I observe that NPC corps often plant agents in multiple systems within one constellation, but ignore other constellations altogether. 5. COSMOS stuff happen in constellations.

Uhmm.. Think it's just human nature to give names to things. So they named each region/cluster/empire so it's easier to talk about them.

The boundries have two significant effects on game play.
1) The boundries allow you to slice the universe into manageable chunks. For example you need a missile launcher but don't want to one that is 10 jumps away. Limit the search to only look at those close by.
2) The prices for products are only quoted by region. Two stations one jump apart but in different regions can have vastly different prices for goods and you would never know. Unless you spent the time physically going to both regions. This gives a chance for traders to make good profits.

Myb undeerstanding is that the backstory is supposd to fill out at some point to make each constellation have it's own significance. Also, it is convenient for me to track my assets by constellation, as I sometimes will have items in so many different systems within one region that it can be difficult to get a concise picture of what I own where.
The game does provide a consterllation and a regional chat channel that is voluntary to join. These channels seem to be never used however.
Finally, I tend to think of regions in terms of constellations. At first, I ignored them and only paid attention to certain market hubs within a region. But I have come to see how different corps and alliances do seem to dominate in various systems/constellations, and how each of these have their own flavor in terms of market and player activity.
Regions do seem to be about racial/empire ownership or player-claimed space. Again, this impacts things such as what npc's to expect in the belts or what racial dependent bpo's and skillbooks are available, and also will have a big impact when factional war comes to the game in the Kali expansion.

I often miss an option to reduce market range to constellation. I really need it sometime, to see whats available in my 0.4 constellation. This, to do trading in and out of the constellaion to 0.5 and up. Also to be able to see... are there any missiles for sale down here? For now, I have to check the region.
