I am a new player and i dont know what is the difference, is 0.4 safer, does it have any defense or there is some other reason why it is 0.4 and not 0.1?

At 0.4 to 0.1 there are gate sentry guns that will defend you if you are attacked by another ship without kill rights to you (who would be concordokkened in 0.5 and above). They can be tanked, and won't do much good to you against a pirate camp that can one-shot you and then warp off before the gate guns finish any off.
In 0.0, you have alliancemates (hopefully) who will protect you (hopefully) and give you intel about hostiles moving about the region (again, hopefully).
I find 0.0 safer, for some reason... At least there you know that anyone you don't know is most likely after your corpse and cargo. Besides, as long as you've got friendly relation to the local alliance and travel rights to the area, you're not going to be harassed very often (again, hopefully).

0.4s are the most dangerous places to be. Very painful experience if you are not careful. Pirate gate farmers like to shoot passer-bys.  ---------------- RecruitMe@NOINT!

If you go deep into 0.0 space, especially off of any "highway" systems, it usually is safer than low security space. Hey, it is safer than some high security systems as long as you keep your wits about you.
Just be paranoid of everybody and get it in your head that everybody is out to deceive and kill you. You'll be safe with an attitude like that.  ___________________

The depths of 0.0 often seem safer than 0.1 to 0.4 simply because there is no one there. You can travel through 0.0 for 3 hours and not see a single living soul even in local. Particularly if you log on from the US during evening hours.
As to the difference between 0.4 and 0.1... Potentially fewer/weaker sentry guns, but I haven't noticed much of a functional difference (I get attacked the most in 0.4 systems and in gateway to 0.0 systems simply because that is where the most people are).

Thank you for your answers. I asked that because i have some trade goods i bought 5 jumps away but on the way i saw a 0.4 system and it has "kill in last 1 hour" and "podded in last one hour" so i guess it is pirate infested :P I guess ill just wait and go get it some other time...or any other suggestions? Of course im in Hauler :)

I disagree with 0.4 being more dangerous than 0.0.
Provided you keep an eye on local and are aligned to warp out to a safespot immediately when somebody even appears on your overview, you're fairly safe in low sec.
0.0 allows the deployment of mobile warp bubbles and interdiction spheres. You usually don't know they're even deployed until you're trapped in one and if you're caught in one, you're normally waking up in a medical clone shortly after.

Is there a link that talks about these "highway" systems or different routes into 0.0 space or is that just something you learn over time.

Make an alt and fly him in there for a look....doesn't matter if he gets podded. You could make some insta bookmarks while you're there and then transfer them over to your main character.

A highway system is a link of systems that go from deep 0.0 hubs to empire. For example, look at Paragon Soul and as you go Northward to Empire space you can see there is pretty much one or two routes that you can take. The other systems, that branch off of these highway systems, is the ones you want to hunt in. People won't see you since you'll be off in the path less travelled, as they say. ___________________

Aye. 0.0 used to be safer but now its not always so :)
as for the OP's question.
0.1s have tougher/bigger rats and slightly nicer ores (or they used to anyway!) and less sentry guns at gates when compared to 0.4s
but a 0.4 is a big step in safety terms from a 0.5 ;)
