Perhaps a few features could be introduced to make regions more inclusive and make our galaxy seem 'bigger'.
Regions as exist now are simply names with all the history being player written and expanded upon with very little in the way of tools to make that happen.
Perhaps an atuomatic chat channel that connects everybody in a region would be a start, from there regional ingame forums and/or ingame station forums would be a big start.
If we are talking about less simple additions we could then postulate that the regions of EVE could use institutions that are player run that are region specific. Perhaps Lonetrek could have a semi-player run organization that deals with the Guristas in events poth AI run and AURORA run. This would largly effect empire space. However, if allainces were allowed to create some further institutions in their areas of control it could affect a new regional community out in 0.0 as well.
The over-all effect of any of the above would be a different and unique atmosphere for every region of EVE. The universe would seem more diverse and larger as people would perhaps develop regional loyalties that currently dont exist.

Its easier than that - just make it impossible to go between the 4 racial 'empires' without going through low sec.
Voila - 4 entirely separate markets, with quite distinct characteristics.

but find a way to prevent everyone form huddling into oen region with all the rest barren wastelands
