you sure can. you can use the search option in the People and Places tab. Just be sure to change it to search solar systems. IT will give you a list of systems that start with the what you entered. right click on the correct system and set it as you destination.

1) Open your People & Places tab, and set the dropdown filter to Solar System.
2) Type the destination. Most times you can get away with just the first few letters.
3) The search-box pops up with system name(s) on it. Right-click the system and select Set Destination. (Or Add Waypoint, if going in stages)
The Set Destination rightclick option is available on more than just this, btw. For example, if you do an Asset Search (need to locate that forgotten Trit?), you can set desinations right from there. Set destnations right from the Market screen, when buying that Mining Laser. Etc.

Is it possible to optimize the auto-pilot route without opening the map? Same reason.

You can also set destinations and add waypoints from your assets window, or from the market interface, and from the missions description window in the missions journal.
You can "optimize" your waypoints by setting destinations and waypoints manually so that the shortest route is achieved... if you know from memory where star systems are without opening the map, uh I guess it's possible.

You'll also find Ombey's Maps useful to print out for reference, if avoiding the ingame map.
