A corp mate and I went into a local belt to test his new defender missiles . . . but as I tossed a few missiles at him . . . he kept getting a "No Suitable Target" message as he tried to block my shots . . . what are we doing wrong . . . it would even give him that message on NPC rats firing at him . . . is there a setting or something set wrong ?

afaik defender missiles dont require a target, just enable the launcher when there are incoming missiles

So ya just turn them on and they activate when a "target" is close . . . a missile I would imagine would activate them ?

Last time I used defender missiles (just before RMR?), you had to launch them manually... but you didn't target anything. Essentially, they worked like FOF missiles, but there had to be a missile in the air (and within range) for you to launch.
For them to really be useful (IMO), they should simply activate whenever incoming missiles are detected and fire at their fire rate until no more enemy missiles are in the air. I mean, you're already taking the penalty of using up high slots to have them, and it takes multiple hits to block the larger missiles (cruise/torpedo)... forcing you to fight network and graphical rendering lag too makes them not so helpful.
Note also, it's not so bad when you're blocking cruise missiles that are coming from 200km away, since you have a good chunk of time to notice them and react. Trying to block light missiles or rockets from orbiting frigates is pretty much impossible unless you're spamming the button.

don't bother trying to get them working. Ever since the patch that boosted missle speeds they are totaly useless. They were mostly useless before that, but at least not totaly.

corp m8s are allowed to shoot each other arnt they ? the defenders probably dont see your incomming missiles as agro
and yes defenders are pretty much broken but i suspect that if you took the skill for defenders to maximum and all the missile support skills they migth work kind of ok

I used them a while in missions, before the missile change that introduced explosion velocity and radius (so that's a long while ago). I could stop heavy missiles quite nicely at that time.
Right now I'd just beef up my tank to receive the extra damage and don't worry about it anymore... Even if defenders would work correctly it would be too much micromanagement to keep it going the way it works now.

after so many posts regarding defenders and CCP and forum officals saying there broke, I'm just waiting for a forum offical to say submit a bug report (Thats not a go at the OP or any other post in this thread)

Corp mates who have fired on me were auto targetted by my FOF missiles.

If you want to fit Defender Missiles at all, do it in a rocket launcher, and really, they're best used on the bigger missiles, like cruise and torp class. If, yes, they'll work for you at all.

The last time I tried Defenders was in a rocket launcher against Zor in his cruise missile spamming Raven, in a level 4 mission.
This guy has a RoF of about 2 seconds. I got in at a good distance to allow my defenders to home in on his CMs, filled a rocket launcher with Defenders and let rip.
End result: 10 defenders killed the leading CM, and after that they failed to kill ANY MORE CMs, because they had less than 2 seconds to home in on the next one and utterly failed to do so.
Improved technique: Ditch the defenders, pile massive damage upon Zor till he stops shooting. Works far better. --

I use two rocket launchers with defenders for missions. Really its just a 'neat' factor, but it does work. Like they said,...ya need time to launch'em. Anything launched at you under 50-60km and they prolly wont catch them in time. Since I'm sitting at 60+ with Howitzers,..they do work. Knocking out a few incoming missles ain't so bad, basically rendering 1 or 2 hi-slots useless of the enemy is not a bad thing.
