Rats have a very low cap capacity, so whacking too much nos on them doesn't work no. It does mess with their tank tho. More nos = less rep frequency. ---

As far as I know, mission rats (or maybe any npcs, not totally sure) have 2 capacitors, one you can suck dry (fairly quickly, hence you gain very little from this) and one you can't touch, so you can't break their tank this way, which together mean that there's very little point in trying to nos them...

NPCs just use a roll-of-dice method to decide whether or not they will repair/boost. Nos only lowers these chances, but will not lower them to near-zero.
Cap gained from the nos depends on the size of the NPC. Battleship sized NPCs have capacitors roughly the size of normal Battleships, although, as far as I can tell, they do not regenerate, so once you nos them dry, they won't give any more cap again.

BS and BCs can get you 2-4 cycles worth, frigs only 1 or 2, is my experience. There does come a point after X cycles where nos wont work on them.

The EVE item database that I cannot find my link to any more listed Nosferatus as having something that affected the NPC's chance to repair/boost shields. I recently dropped NOS from my mission ship and think they repair a bit more often, but I haven't really added up numbers.
Also listed was something about cap boost on the NOSsing ship, and my cap is a bit worse now. I usually did not switch NOS targets but kept the beam on one until it popped, so I doubt the NPCs give a limited amount of cap but instead a fixed small number. But again, no hard facts just my impression.

There is something I do that requires me to NOS NPC to attain a sustainable tank, so I'm pretty familiar with the effects of NOS on them, depending on their cap to run my tank.
It is true that after a number of cycles, they are dry, however switching NOS between targets and coming back to NOS the dry ones again will give me cap again, so they do regenerate their cap. They seem to take on average 5 minutes to recover, and their ship size or ship type affets their cap size, for example a NPC blasterboat gives me more NOS cap than a ranger boat and the blasterboats seem to recover NOS faster too.
