Do you mean if you are using them? If that's the case, charge up to your target (or into the middle of multiple targets) and activate. That's about it.
If you are fighting against them, try to stay out of 5km from the enemy ship. There are some smartbombs that will reach further, but they are expensive.

Asestorian has the right of it. Smartbombs hit anything in range (5km for the large ones...further for very expensive faction stuff). No need to orbit or anything. Just be in range and done deal.
Be very careful with their use in hi sec space. If someone who is not a legitimate target is in range and you hit them (or even some structures) CONCORD will kill you. Most people find it too dangerous to themselves to use SBs in hi sec for this very reason. You can, just be careful.

Does it matter in missions? Because it looks like I've blown up many structures as collateral damage and hit even acceleration gates can never saw concord anywhere.

Smartbombs have very limited, specialized, tactical uses. I definitely wouldn't use them for missions. ------------------

Some structures, both for story wise and for strange reasons (I suspect the devs forgetting to set them to hostile) are considered concord protected in missions. If you use smartbombs in missions be absolutely sure that any structures in the blast range are hostile.

Speaking of smartbombs, I've hurd rumors from various mudkips that they will take down missiles in range. Is dat troo?
It's kinda hard testing it, since not many missiles spend too much time within a 5km radius from you.

Yes it is true but as a missile defense it is very poor. Due to lag issues (always some lag present even if all seems fine), missile velocity to cover those 5km, cycle time and so on you'd mostly be lucky to kill a missile with a smartbomb. And for the one you kill you spend a huge amount of cap and get hit by the next 20 missiles.
Go out and test and you'll see what I mean.
They are pretty good at killing drones though.

Poor as they are for missile defence, they still work better than defender missiles...

Interesting. I was curious because that would allow me to intercept missiles aimed at other players. When my friend does missions in his battleship and we end up fighting caldari or something, there's a huge torrent of missiles heading his way. It would be fun trying to smartbomb those.
Though I guess simply remote shield boosting him would be more functional.

Shield boosting would be far better.
Remember a smartbomb hurts everything within its range...including your friend. If your friend is not a corpmate (player corp) and you tag him in hi sec CONCORD will kill you.
Beyond that you'd need a lot of smartbombs to be even remotely effective and that is not an easy fit and they positively wail on your cap so you are not likely to be able to keep it up long.
But have fun testing it...just be careful about what is in range of it and possible CONCORD issues.
