To clarify, both target painters will work, the second one won't be as effective as it should be.
For example, two 30% target painters would give a (30% * 26.1%) 61% bonus rather than a 69% (30% * 30%) bonus.

As an aside, I'm a big fan of target painter. I use one instead of a web stasis on my ratting Domi, because I can't be stuffed to go chasing NPCs just to get into 10Km range for the stasis. -

Note that once you increased the ship's signature to that appropriate to your weapon (increased it to cruiser size if you are using medium guns or heavy missiles for example), there is no more gain to be had from additional target painters. They are all but useless against size appropriate targets. |

For missiles, yes, not for turrets though.
Assuming the Tracking formula is correct or near enough, the chance to hit improves as the signature resolution/signature radius term becomes smaller. There is a limit to its effectiveness, mostly defined by diminish returns, but increasing the ship's signature radius is always a good thing where turrets are concerned.
