I know nothing about using player couriers.
I know you can only place the contract at the station the items are located. If I have 50mil in goods to move, I am assuming I ask for collateral in the contract.
How much do you pay out based on? Volume? Value of total items being moved û represented in collateral? Number of jumps?
Is their a typical % that you provide based on the total collateral or number of jumps?
Example û if I wanted to move a large volume (80,000) of high value items (totaling 50mil) 5 jumps, what is the payout? The same contract but 10jumps?

You were right the first time, next time stick with your gut.
You can remotely create courier contracts, just go to your assets, right click and select contracts then courier. It works fine with private contracts, I haven't tried public ones.

80,000m^3 of items for 50 mil is not considered high value items. That's very low value. I often move single items worth more than that.
If it's a low volume but high value I'd say .5% of collateral is a good idea, minimum of 500,000 isk or so from my experience. I pay 500k for anywhere from 50 mil to 150 mil worth of low volume items. I pay more for higher volume. And when I say low volume I mean 1-100 m^3. If it is 1000+ you have to pay more (it will be done in an indy, not a frigate) and if it is over 15000 you may have to pay more as well, especially if it is over 30,000.

You might want to check out EMMA (link in my sig).
From there, as long as you have enough data, you can generate courier contracts automatically where the collateral can be a specified percentage of purchase price or expected sale price (I use 105% of sale price).
The amount of reward can also be specified by different parameters. For example, I tend to calculate reward based on estimated profit. minimum will be 8% and max will be 25% where 2% of profit is added per jump and 8% is added for a lowsec pickup or dropoff.
Currently, the main limitation is there is no way to include the volume of the contract in the calculation and this is very important. So if you have a particularly large contract I'd perconally add a little more.
My rates are well known for being rather high generally though so if you don't need really fast delivery or if you really care about saving every 0.01 isk then you can easilly get away with lower values.
