Is there any kind of best or optimal region to trade in? I have 30 mil ISK at the moment so I have more than enough to trade is most any good.
Also as a side question, i'm using a Bestower ATM with 4 cargo expanders... what would you put in mid slots that would help with trading aside from an AB? Also i'm using this eve central trading tool in the IGB, is there anything better than this for trading?

There isn't really a "best" region although it seems most serious traders will operate around Jita (THE trade hub of Eve).
For your indy it's generally a good idea to put on some shield extenders in your mid slots if you have the grid if you're going to be moving goods in it on a regular basis as these will give you a slightly better chance of surviving a suicide gank (no guarantee but better than nothing).
Finally you won't get many useful answers on what to trade in as the traders themselves aren't going to encourage competition by handing out trade secrets. Your best bet is to work it out on your own by just playing the game really.

The best regions for trade really depend on what you trade, and how you like to do it. People will often cite Jita, but that's not necessarily where the best prices are for a seller. For the things in which I deal, Jita's the last place I want to be...not to mention the lag.
The bottom line is, you have to do some homework. Trading is the biggest sandbox of them all. No one can tell you what to trade, how to trade it, or where to do it. Don't expect people to spill their secrets to you, either. They did their homework, and sharing that info is sharing their bottom line with you.
On a side note, is there any thread now that doesn't mention suicide ganks? Lol, it really shows you what the flavour of the month is
