A tank is nice, but the best way to keep from dying is to lower incoming DPS as fast as possible. To do this, you need to fit some megastabs to increase your DPS.
Also, aggro control is important. Things are much easier on your tank, when the whole room isn't shooting at you. Make sure your drones are on passive, so they don't aggro a group of rats you don't want to attack yet.

For an ubeatable mission tank, go with dual large reps. One hardener for each damage type dealt, so for Serpentis, use a thermal + kinetic + EANM (buffs both). This set up perma runs without the guns going, and lasts for 10 minutes with them on, plenty of time to kill everything in the mission.
does that include guns?

High DPS is really what you need for missions <.<
Always a 1 x LAR, 3 x Hardeners + Damage modifiers are a much better tank then a ridicolous dual-rep, 4-5 hardener tank.
Kill all that incoming DPS and you're fine, also you will actually make more money missioning then high-sec mining
