The absolutely quickest way to make heaps of ISK (legally) and with the least effort is to buy a GTC from CCP and use it to create and then sell two PLEX's.
It will take an hour and you will be approx 600M ISK richer.

I meant with out spending actuall cash, like working in game to make ISK.

well that really depends on your skills.
if most of your skills are in mining....join 0.0 alliance, or use wormhole and mine mercoxit or ABC ores.
if your skills are in combat Level 4 missions(or work up to it) or rat in 0.0 or sleepers in wormholes.
or exploration (archelogy salvaging)
if your low skill level probably the quickest cash is train destroyer and salvaging...and find someone you can follow and salvage their missions, or salvage anomolies in 0.0.
Trading (requires a little nest egg to start) can work but usually requires a lot of babysitting the orders.
The better question is what do you want to do....if you just try to make ISK the game gets boring quick.
The answer you want really varies....a 2million sp character would get a different answer than a 5 million or a 10 million or a 20 million sp character.

I could give you the answer but where is the fun in that :) Seriously part of the fun you can have in eve is figuring out how to outsmart the other chumps grinding for isk. There are loads of ways. Here are some clues. 1. mining for a new player is not the most profitable thing to do at all. 2. the game eve has a very well developed item exchange system aka the market. The devs putting so much effort into it should give you a clue. 3. ? 4. profit

For a new player, the most profitable way is to do missions (aka mission running). eve-survival.org is a website that can help you a lot because it lists everything you need to know about every mission that you can possibly get from an agent. Doing level 4 missions is one of the most profitable things you can do in the game, but you must get to Lvl4's through lower level missions.
The quickest way is trading. EVE is based on a real world economy. That means that not many items are "seeded" by the developer of this game (that would be CCP). Almost everything in game is player produced. To be able to trade in EVE, however, requires that you know what you are trading with. You must know your stuff what's it used for, how rare it is, how you can get/manufacture it, are there more useful variations of modules of the same type, what types of players are using those modules you are trading with etc. Trading would also require some ISK so you can invest in your initial stock of goods, so it's not generally a good option if you are that new to the game.
Mining can make you rich if you have a couple of friends (with at least one Orca pilot) and you are flying a Hulk in lawless 0.0 security space. The most profitable ore is in 0.0 space: ABC ore (Arkonor, Bistot and Crokite) and Mercoxit.
Since making isk in EVE can be done in many ways and each profession can be very profitable, I can't list all of the options here. Instead, take a look at http://wiki.eveonline.com/en/wiki/Making_ISK and continue exploring that site to get familiar with the game (wiki.eveonline.com is an official CCP's guide web site).

Scamming and trading, even better is some combination of the two. There really is no easier way to make stunning amounts of ISK.
Mining in high sec is terribad ISK compared to the other carebear acitivities. The safest ISK faucet, and the most lucrative, is to find a solid quality agent in .5 space, with a corp that has a highly desired LP item or 3. Ideally, you want to get a 2,000 ISK conversion or better. Next, max out your social skills to farm LPs as quickly as possible. Train up a beastly Lv 4 mission grinder, and have at it. Ignore ALL loot and salvage unless you've an alt on hand who can work along behind you. Within a very short amount of time, you'll be making extremely good ISK, leaving you ample time to do whatever else you find interesting.

If any such thing existed (something being CLEARLY and universally the quickest and most efficient way to make ISK), everybody would start doing it instead of just about anything else, and it would cease being the quickest or most efficient way to make ISK. The quickest ways to make ISK are either the least efficient, requiring most time/work/attention or are extremely risky... and the most efficient ways to make ISK are either not quick at all or require massive initial investments (or both). So no, you can't eat your cake and still have it afterwards.
TL;DR : try a bit of everything described in there and settle on what works best FOR YOU personally.

I would highly recommend avoiding mining. There are far better things to be doing with your time and your character's SP.
While you're reading that Making ISK guide, start mission-running to build NPC corp & Empire standings. Standings will be useful for you regardless of your future career.
Make friends. With a group of friends flying cruisers or battlecruisers you can take on level 4 missions and be making much more ISK/hr than level 3 missions while having an absolute ball of a time.

Yes, 0.0 rats in asteroid belts can make you a decent amount of cash, especially if you exploit the spawning system to chain them for higher bounties.
However, I would not recommend this for a new player without experience of low-sec/0.0, you need to know how to survive should any bad guys come along.
Wormholes can be pretty profitable as well, but these are even more dangerous than 0.0 in many ways, and many lower-level WHs are now occupied. You also need to consider that you might need to go back for a dedicated salvage ship to loot effectively, and consider the danger of getting trapped should your exit hole close.
If you're new, build up to L4 missions, get a salvage boat as well, and then get some tips on how to survive in more dangerous but lucrative parts of space.

There is only one quick way to make lots of ISK for a newbie - it's called scamming. All others require some knowledge, time and patience.

As I was saying, there are various "named" NPCs, called "officers". They spawn only in 0.0 space, but the loot they drop can be worth *billions* of ISK. I named "Estamel" specifically because Estamel has a chance of dropping "Estamel's Modified Invulnerability Field", which is perhaps the most valuable module in the entire game, worth even more than most capital ships. ----------

You probably could have been getting into one of those big-ass ships for a long time before you have a good chance to encounter Estamel even once. And no, you don't need a capship to kill it, a single battleship will do too. Heck, a single battlecruiser will suffice, if properly fitted. There are L4 mission mini-boss NPCs that are only slightly less tough to kill.

I'll recommend missions.
Missions are decent money, but more importantly, the skills you put your skill points into are mostly never a waste.
Even if you abandon mission running later, you'll probably keep using "most" of the skills you trained.
There are very few things you can end up doing in Eve where you are completely un-interested in having good ship fighting skills on a character.
(Having said that, some skill points are almost always wasted while you are figuring out what you want to do. Don't lose sleep over it. But ship flying is useful to 'almost' everyone....) Play nice while you butcher each other.

From a player who started at the beginning of June (still relatively new), mission running has easily been the best source of income. That being said, I don't just run missions, I 'clean' them. I run in a drone boat, Domi set up for tractors/salvs, and 1 gun to get aggro. Salv/loots/refining/selling add up to a significant source of my income. I plexed this month, and with ~20 days left until this one expires, I'm already at about 150m.
I certainly won't clear a mission as fast as more experienced players (and only having played a few months I wouldn't expect too), but there is money in nearly every aspect of Eve, you've just got to squeeze out every isk you can.

Reprocessing loot items is another method of making isk.
Some loot drops are worth 2-3 times as much as reprocessed minerals as the buy orders on the market. Meaning a 2 million isk buy order can be worth 4-6 million isk as reprocessed minerals.
The best part, you can set your buy orders close to what the sell orders are doing and still double your investment. All you have to do is take a couple hours once a month to pick up/reprocess your items. All it takes is a little research on the market to figure out what items are worth setting up orders for.
