Hey guys, I'm trying to figure out how much tank do I actually need according to NPC Game data and EFT so I can fit the most DPS. Lets take a look at the mission Worlds Collide, Guristas v Serpentis.
The Guristas side does 1,210 Kin & 299 Therm DPS. With my 82% Kin and 76% therm res they should be doing 244 DPS total to me. If make a damage profile of that on EFT it comes out to say 414 under defense. Does that 414 defense mean that I can tank 414 DPS? or is that just a random rating?

Yes, it means that you can tank 414 DPS on that damage profile, if it is correct. I myself did WC serp vs guri (not other way around though) in a drake ~300 DPS EFT tank (common serpentis damage profile), but it's with an AB fitted (speed tanking with about 300 m/s is kind of funny but in fact it really helps against npc BSs).

EFT doesn't tell you the whole story. In this case, what it's telling you is that, if you draw full aggro and just sit there not shooting back, you'll die in a fire. How long it will take is determined by how large your buffer is, compared to the incoming DPS that's above your ability to tank (in your example the difference is ~1100 DPS).
But there are other factors: Can you manage aggro so you won't have to take all 1500 DPS? Can you kill the rats fast enough to reduce incoming DPS to bearable levels before they chew up your buffer? Can you get moving fast enough to significantly reduce incoming damage through "speed tanking"? All these factors can have an enormous effect on the real amount of tank required.
Also, don't rely on available NPC damage profiles to be all that accurate. The data you're looking at is likely to have been drawn from an old database dump. It's information that CCP doesn't supply any more, and so is less and less reliable as time goes on (other than percentages; Guristas will probably always do almost entirely kinetic damage).

Also, it is more than just total DPS that you have to consider. For example, I am rather fond of using the Arbitrator on level 4 missions in nullsec or lowsec when there are people around because it is a cheap loss if I got sloppy, but I don't use purger rigs on it I have extenders, in fact my EFT claims a sustained defense of only 95 omni with a real HPS of about 48. The reason why is basically you only need to tank average DPS everything beyond the average is limited to the buffer your ship has.
So, for example when I enter the second room of worlds collide 4 Angel/Amarr/ammatar*, the ammatar frigs and cruise battery will autoaggro (the blood one had no missle batterys but it had autoaggro with frigs too so I am assuming yours does too) and it will do more damage than I can tank, however, I can kill the webbing frigs without drones reasonably quick, and then with my speed back it is very easy to pull range and kill everything while letting my shields regen.
As far as to the numbers thing 414dps is what you can tank on your damage profile, I downloaded the custom ones that have the same values as the factions average, but as the other posters have said you will only tank that standing still, an AB will help out a bunch so it is hard to say how much abuse you can really tank, but I know I never had an issue fitting 3 BCS II on my drake and still tanking missions, though that might depend on your skills. I am fairly certain it is less than 200 sustained, but has plenty of buffer on my drake.
*The mission says against amarr, and the first pocket rats are ammar, but the ones after are ammatar

I've found that if you have around a 500 - 600 dps tank versus the faction you are fighting for that mission you are fine. That's the baseline I use if I'm not speed tanked.
400 dps and you may occationally need to warp out, 700 dps and you are way overtanked. 600 will let you sit in a room and pull everything. Keep a damage control on your boat if you are not sure, it will give you time to jump out if you find you've overpulled. =============================== Two words: Internet Spaceships |

Thanks for the info guys, I have been taking missions with about 400 or so fine for a long time, only warp out when I forget the change my hardeners sometimes. I was just wondering cause I wanted to get more damage mods without actually dying.
