Every time I've done 16 missions of a given level, I get a storyline mission.
But let us say that I fly 16 level 2 missions, and then I get a storyline mission EVEmail. I then fly 2 more level 2 missions, before I fly the storyline.
How many level 2 missions do I then need to fly, before I get another storyline mission? 14 or 16?

should be 14. If you do enough missions you can queue up storylines - you'll just get them from the next closest available agent.

I think you need to do at least one mission after a storyline to draw another storyline. Someone please correct me if I'm wrong.

You can queue up a lot of storylines, at least as many as storyline agents there are in your region. Never tried queuing up more than 3 storylines though. Each storyline will come from a different storyline agent, and queuing them up is a great way to select which corporation will you receive the huge corp standings bonus with (only take and complete the ones from agents you don't like whenever they're close to expiration date).

You can also pick which storyline agent will give you a mission more directly. Just do 15 missions with any agent you like then carefully pick the 16th so that the storyline agent you want the offer from is the closest storyline agent to your 16th mission.
