Is it possible and easy to maintain good standing with all the important non-pirate NPC corps and factions/races while doing missions for agents?
For Example will Caldari agent missions ruin standings with Gallente?

"Good" tends to be subjective.
You will not be able to maintain high standings with all factions: high being enough to allow to anchor a high-sec POS in all regions.
You could potentially maintain modest faction standing (enough to access most level 3 agents), but it will require you to keep switching which faction you are working for.
Keeping all of your base standings postive is not too hard. It would require a moderate amount of balancing, but it shouldn't be too bad, maybe changing factions every month or so.
Keeping your effective standings postive is easy as Diplomacy makes a huge difference. You might need to switch factions once or twice a year, if that.
Keeping them high enough so that you don't lose access to running missions for any of those factions, with Diplomacy V, only requires you to turn down any missions that send you out to kill the empire faction NPC's.
With Diplomacy V, you would need to put a significant effort into hunting empire NPC's before you would become KOS.
It is really up to you how balanced you want to keep your standings. From my experience, simply turning down (or limiting how often you accept) missions against the other empires, does more than enough to keep your standings reasonably balanced.

If I lose all chance of even a level 1 agent even with Dipl V. Can I get it back by grouping with someone else doing an agent mission and thus sharing the benefit? If not then how?

Nope.
Fleet completion of a mission will allow you to increase your corp and agents standings, but not your faction standings.
If you have an effective faction standing of -2.0 (base standing of -5.0 with Diplomacy V) then you will not be able to run missions for that faction, regardless of how good your corp and agent standings are.
The only way to increase you faction standing is by running missions for that faction or its allies, or by earning Faction Warfare ranks with the same.
So if you nerf your standing with Minmatar, you could run missions for Gallente, SoE, or ORE to repair your standings.
If you have nerf'ed your standings with all of their allies, then you can repair your empire faction standings by joining a player corp in that faction or its ally's militia and earning ranks to gain standings: this is a one time option as you only get the standings increase the first time that you earn the rank.
With pirate faction blocks, once you cut yourself off from a faction and its allies, you can not repair your standings under current game mechanics. That might change in the futre, but there are not yet any specific plans for such a change.
