It is a pretty serious answer. Those who get rich hardly want to give away their secrets to potential competition.
However, rich people may have done it through trading, POS networks, stealing, scamming, suicide ganking, manufacturing. All kinds of things.

CriticalHurt, your best bet will be to find a niche. something nobody else is doing.
i made half a bil in a week buying the "Armour Plates" salvage item for 1 isk a pop when it wasn't used in the production of any rigs. when that was patched i dumped it all the next day at 45k per unit, which was relatively cheap but netted me a massive profit without any hard work at all. i didn't even need to think about ratting for a couple of months.
holding out would have netted me more cash, because the price rose after the stockpiles the mission runners had been keeping ran out, but i'm not greedy and fast cash is where it's at.
that's just an example, and there were a few of us doing this so already people will be looking out for the next 'useless' item CCP are planning on giving a purpose. I have a sneaking suspicion that at least some of the people buying corpses in bulk are hoping they'll be able to do something with them in future.
just look what happens to ship prices whenever a buff is rumoured.
the other thing is day trading. buy low, sell high. lots of people doing it though, so make sure you know your ****. my last foray into day trading left me with a few hundred units of t1 resistance plating i don't particularly want.

Trading seems to be a good way since it is the only activity I found makes you money while you are offline.

Hmm - well, if you send me 100m isk, I'll tell you how to make money from gullible idiots 
If you're lazy but well-informed, you can earn quite good margins on a large amount of isk by gambling on how the value of certain items will change. Ideally, spend plenty of time on the test server, and look out for any interesting changes.
I can go one better than ry ry - I bought up several copies of the Astronautic Engineering skillbook when it was accidentally released last year and sold them to collectors at a 900% mark up. Granted, some people made a lot more isk that way than I did, but it taught me an important lesson - people are prepared to pay an enormous amount, even for complete junk, if the only way to get it is to pay an enormous amount.

If you are into trade and such activities, head over to the Market Discussion forum and check out some of the threads in the Essential Thread Sticky. I myself am not very rich as each time I have enough isk to buy a new toy I do and then promptly get it blown up, however, I have known many people in this game that have made billions in relatively short order by just running lvl4 missions.

There is hundreads of ways to make isk in this game. Mining and ratting have always been the slowest ways to make isk though. But there is a tried and tested method that has worked countless times for newbies.
Buy t2 pvper gear in jita.
Sell in warzones or in highsec nearist to hotspots (such as agil/Torrinos etc etc).
Do that till you get a few billion, use transport ship and never carry more than a couple hundred mill worth of stuff afk
From there, you will have seen enough of the game to know where to start creating oyur own supply/demand etc.
Or just run lvl4's. Whatever works for you.

create a new galente>Jin Mei>highest perception possible (saan go caste? i think)>military>soldier
Then train up weapon upgrades 1, destroyers 1, and electronics 1.
Buy yourself a shiny new gallente destroyer, and fit it with 8 small ion blaster 1's(filled with 6 caldari navy antimatter each), as many MFS's as you can fit, and a cargo scanner. Then get a few of your buddies to do the same, and get one person with a hauler alt. Then just sit off the jita gate scanning afk haulers, or on a trade route scanning autopiloters.
It takes some time, but every now and then someone autopilots a hauler full of zydrine, or a shuttle with an officer mod, or something like that, and you hit the big bucks. When we first started we were pulling in 200m drops an hour on average.
Also, for less money but higher probability of payouts, do the same thing vs highsec hulk pilots. They're usually quite expensively fit, and sincey our ships cost next to nothing anyways, its easy money.

I won't give out any simple tips but instead a thought process you should have.
Where is the money?
Who wants to buy/sell what and where? Who carries what and where? And more importantly, how can I get the stuff from them or get stuff somewhere so I can sell it instead of some other random guy?
If you think about, say, minerals: where someone is mining for them? Where can I buy them cheaper and in larger quantities than elsewhere? Where do people need them? Where should I sell them to maximise my profits? How can I make sure that nobody disrupts my business?
Think what you want to do, try to find out a way to make the most money out of it. If it doesn't earn you much, switch fields or improve.

Which means you can't have any fun flying ships and all that, since that costs isk.

I've found the best way seems to be by amassing an ungodly amount of ISK. Some prefer exotic dancers, though.
