I'd have that PC checked if I were you. there's definitely issues if none of the audio I/O jacks work.
The first solution that comes to mind is replacing the mobo. and no, I'm afraid you won't be able to avoid paying money to fix this issue.

For anyone capable of puzzling this out, I was on Mumble last night getting ready to record. Everything was working fine. Kyle thought about trying to record on Skype since most of our podcasts lately have been troubled with cracks and pops. I closed down Mumble and the call was made on Skype.
I couldn't hear anyone. They could hear me.
I tried rebooting my computer. No change.
I relocated my headset from the front jacks to the back. No change.
I relocated them to the front, and now it doesn't even acknowledge that they've been plugged in. No mic. No sound.
I tried my kids headset (my old set) and the same.
Needless to say, we didn't record last night.
I'm using an ASUS crosshair V Formula Z motherboard.
Does this sound familiar (like a cheap easy fix)?

That particular board has some way of setting up sound that baffles me to this day. When you plug in speakers or mic's, a window pops up acknowledging that "something" has been plugged in. It asks for confirmation.
Thats where I knew I was having trouble last night was when I moved the headset plugs, no window popped up. It's almost like the board doesn't acknowledge that they've been plugged in anymore.
Like I said, I know that I'm asking for crystal ball diagnosis, but I'm just wondering if these things give an indication of where to look.

not sure there mate , mine i can test , i use a sound card so i can test , speakers , headphones , mic so i can trace the fault , if your running windows go in thru the control panel and check your sound options , you may be able to test it that way , you may have a key that defaults the mute button too, iv'e done that, accidently muted mine and nothing works
